Cytonome demonstrates rapid and sterile regulatory T cell purification

Cytonome, a developer and manufacturer of advanced cell purification systems, today announced a milestone in the rapid, sterile purification of regulatory T cells. The study, published online in the journal Cytotherapy, describes the use of the Gigasort™ cell purification system to isolate regulatory T cells from a sample of mononuclear cells. Regulatory T cells are currently being tested in clinical trials for the treatment of autoimmune diseases (such as diabetes), graft-versus-host disease, and transplant rejection. Purifying viable regulatory T cells in a clinical setting has been a major challenge, due to the rarity of these cells and the need to purify them in a sterile environment.

In this study, Cytonome researchers were able to purify up to 1.7 million viable regulatory T cells from 450 million fresh peripheral blood mononuclear cells in under 4 hours using the Gigasort™ cell purification system. Cytonome’s Gigasort™ system uses parallel sorting technology coupled with a closed, sterile disposable cartridge to provide increased throughput, safety and sterility in applications such as regulatory T cell production.

“Sterile purification of regulatory T cells is an extremely important step if we wish to use these cells in a clinical setting to help organ transplantation patients. In our study, Cytonome’s Gigasort™ system was able to produce a clinically-relevant number of sterile and pure regulatory T cells with excellent viability. This study demonstrates the feasibility of using this technology in future clinical transplantation studies.” according to Dr. David Berglund, a co-author of the study and a principal investigator in transplant immunology at Uppsala University.
